静电纺丝-微流控芯片提高艾滋病毒检测效率
近日,国家纳米科学中心蒋兴宇课题组在纳米技术与重大疾病早期诊断方面取得新进展,相关结果发表在最新出版的《先进材料》杂志上。
早期准确快速的诊断是发现并控制重大传染性疾病(艾滋病、禽流感、乙型肝炎等)的必要条件。预防人类免疫缺陷病毒(HIV)目前仍然没有有效的疫苗,抗病毒治疗也不能有效的将病毒从体内清除,并且现有的HIV确认试剂盒诊断所需的时间较长,价格昂贵。因此,开发有效的、高灵敏度快速准确地诊断HIV感染者的检测方法,可以有效防止病毒的继续传播。
现在对于HIV的检测其技术原理主要是基于蛋白质(抗原或抗体)之间的相互作用。微流控芯片技术具有制备简单、试剂用量少、操作方便等优点,因此在生化分析中的应用越来越受到重视。
蒋兴宇和其博士生仰大勇与中国疾病控制中心性病艾滋病中心的马丽英、邵一鸣合作,采用静电纺丝技术制备纳米纤维薄膜,应用于微流控芯片,检测HIV。与商业薄膜相比,静电纺丝纳米纤维薄膜具有更大的比表面积,对于被检测物的吸附提高了一个数量级,从而使得检测的灵敏度有很大提高,在一个小时内就能完成检测工作,使用的试剂为常规用量的几十分之一。
这是一项将纳米技术应用于疾病诊断领域的成功例子,该工作开辟了静电纺丝应用的一个新领域,同时这种结合微流控技术和静电纺丝的新芯片系统具有廉价、操作方便、便携、灵敏度高的特点,将推动重大流行疾病早期诊断的研究和产品开发。
上述工作得到了科技部、国家自然科学基金和中科院的支持。

Now for the HIV testing of its technical principle is based mainly on the protein (antigen or antibody) interaction. Microfluidic chip technology has the preparation of simple, low reagent use, easy to operate, etc., so in the biochemical analysis of the growing attention.

Compared with commercial films, films electrospinning nanofibers greater surface area for adsorption was detected objects to improve an order of magnitude, making the sensitivity of detection has greatly improved, in an hour will be able to complete the testing work, reagents used for several one-tenth of conventional dosage.
This is a nano-technology will be applied in the field of disease diagnosis of successful examples, the work opens up applications Electrospinning a new area, and at the same time this combination Microfluidic Electrospinning technology and a new chip system is cheap, easy to operate , portable, high sensitivity, will promote early diagnosis of major endemic diseases research and product development.
