HIV导致免疫系统B细胞耗尽

  最近的研究表明,HIV可引起免疫系统产生强烈和持久的免疫应答,最终导致关键的免疫系统细胞-CD4+和CD8+T细胞枯竭,CD4+和CD8+T细胞是机体抵抗HIV的主要免疫细胞。精疲力竭的免疫细胞抵抗HIV的能力越来越低,最终衰竭的免疫细胞失去对付HIV的能力,丧失清除体内HIV的能力。



  现在美国国立过敏与传染病研究中心的研究表明,相似的衰竭现象也在免疫系统另外一种细胞中出现,那就是分泌抗体蛋白的B细胞。



  大部分没有接受抗逆转录病毒疗法的艾滋病患者体内HIV病毒持续的复制,导致全身多组织器官功能紊乱,其中保罗引发血液循环中的B细胞亚类发生改变。其中的一个亚类是组织样记忆B细胞,这类B细胞在没有接受抗逆转录病毒疗法的HIV感染者体内含量特别丰富。这类B细胞表现出早熟和低抗HIV的能力下降,分泌抗体的能力下降。



  HIV感染者除CD4+和CD8+T细胞衰竭外,新的研究结果表明,B细胞也衰竭,这就是证明HIV能损伤机体复杂的免疫系统,我们治疗的挑战就是要重建或者是支持HIV感染者的免疫系统。



  美国国立过敏与传染疾病研究中心的疫苗研究项目的目标是,加强对B细胞的研究理解,促进疫苗的研发,这一研究就是为该目标做出了贡献。研究者希望能设计一种治疗性的疫苗,减缓HIV的疾病进程,使失效的B细胞恢复分泌高质量抗体的能力。
